“Toxoplasma on My Mind (and in my brain): Tales of Life Cycles and Cell Cycles”
Seminar Abstract: The Toxoplasma life cycle contains both a sexual and asexual phase. The asexual phase manifests in all warm blooded vertebrates and is characterized by the actively replicating tachyzoites in the acute phase and the “dormant” bradyzoites in the chronic, life-long phase of the infection. We recently challenge the prevailing dogma in demonstrating that bradyzoites within tissue cysts are capable of considerable replication in vivo. In the course of investigating the bradyzoite cell cycle we focused on the role of a family of deubiquitinases (DUBs). One cell cycle regulated member of the Ovarian TUmor (OTU) family of DUBs, TgOTUD3A revealed a remarkable phenotype following its disruption. This mutation results in the dysregulation of the cell cycle architecture such that close to 40% of parasites alter their replication strategy to deviate from producing 2 progeny per cycle to generating multiple (3,4,5) progeny per cycle. As such this dysregulated mutant exhibits replication strategies observed in the initiation of the sexual cycle, and also represent the strategies employed by other Apicomplexan parasites like Plasmodia and Sarcocystis spp. Together our findings suggest a regulatory nexus governing life cycle transitions with the plasticity of cell cycle architecture in Toxoplasma.
