Abstract: Despite recent advances, the high-resolution analysis of diluted organic molecules and biomolecules in solution remains challenging. While liquid-state NMR spectroscopy is in principle able to meet this challenge, the wide applicability of this technique is hampered by its extremely low sensitivity. We developed low-concentration photochemically induced dynamic nuclear polarization (LC-photo-CIDNP), a powerful optically assisted NMR technology for the hyperpolarization of small molecules and proteins in solution. In this lecture, recent advances and applications will be discussed. Remarkably, LC-photo-CIDNP enables rapid NMR data collection in the low micromolar-nanomolar concentration range. 1D and 2D NMR spectra of neurotransmitters, aromatic amino acids and proteins can be readily acquired at high applied magnetic fields and on benchtop NMR spectrometers acquired at high applied magnetic fields and on benchtop NMR spectrometers. The LC-photo-CIDNP technology works well in both pure solvents, buffered solutions and highly complex physiologically relevant media including cell-like milieux. The above advantages pave the way to a variety of applications targeting the hypersensitive high-resolution analysis of molecular structure and dynamics in situ in the context of organic chemistry, biotechnology and medicine.
